Molecular cloning of exons II and III of the a-globin major gene from Odontophrynus americanus 2n and 4n (Amphibia, Anura) Braz. J. Genet.

The <FONT FACE="Symbol">a</font>-globin major genes from diploid and tetraploid Odontophrynus americanus were studied using PCR-based technology. The cloned and sequenced amplified fragments were shown to contain most of the exon II sequences as well as the whole exon III sequence of the <FONT FACE="Symbol">a</font>-globin gene. Unexpectedly, intron 2 was entirely absent in the amplified fragments of both 2n and 4n origin. High conservation was observed among the obtained sequences when compared to corresponding sequences from human and Xenopus laevis origin. The possibility that these sequences might be pseudogenes is raised

First description of microchromosome in the Anostomidae fish Schizodon nasutus from Argentina Braz. J. Genet.

Thirty-six specimens of Schizodon nasutus (Anostomidae-Characiformes) from the middle Paraná River (Posadas, Argentina) were analyzed cytogenetically. The karyotype of this species was similar to those described for this species in the literature. C-banding technique showed a rich heterochromatic pattern relative to other Anostomidae species. The NORs were located on one chromosome pair in terminal position and showed a very marked size heteromorphism. A microchromosome was observed with a frequency of about 20% in the sample studied. This additional element was punctiform, negative C-band, and constant in all metaphase plates of the seven carriers. Cytogenetics of six Brazilian species of Psocoptera Braz. J. Genet.

The karyotypes of the following six species of Brazilian Psocoptera are reported: Caecillius sp. (Caecillidae), Triplocania ? caudata New (Ptiloneuridae), Brachinodiscus cf. lepidus (Banks) (Psocidae), Psococerastis interrupta New (Psocidae), Ptycta nr reticulata New (Psocidae) and Trichadenotecnum sinuatum New (Psocidae). All of them had males with 2n = 17 and an XO sex determining mechanism.

Callus induction and plant regeneration by Brazilian triticale and wheat genotypes Braz. J. Genet.
In order to determine the in vitro behavior of Brazilian triticale, 16 triticale genotypes, and three wheat genotypes used as checks, were sown in June 1994. The explants used were immature embryos. In addition to the genotype tests, two culture media for callus induction were also evaluated, i.e., MS (Murashige and Skoog, Physiol. Plant. 15: 473-497, 1962) medium containing 2.0 mg 2,4D/l, and MS medium containing 4.0 mg 2,4D/l. The plant regeneration protocol used was the one employed at the Laboratório de Cultura de Tecidos, Departamento de Plantas de Lavoura, Universidade Federal do Rio Grande do Sul, for wheat. New thermal inducible Streptomyces phages isolated from tropical soils Braz. J. Genet.

Two new Streptomyces phages, øBP1 and øBP2, were isolated from tropical soil samples. These phages presented a large host range and developed both lytic and lysogenic responses in different Streptomyces species tested. Variations in the incubation temperature showed to be important in the development of the replication cycle. Increasing incubation temperature from 30oC to 42oC induced the lytic response of øBP2 and lysogenic of øBP1 in the host strain Streptomyces sp. WL6. øBP1 and øBP2 have icosahedral heads with long tails and were characterized in relation to morphology, G + C content, genome size and adsorption curve

Using the best linear predictor (BLP) in the selection between and among half-sib progenies of the CMS-39 maize population Braz. J. Genet.
Data of corn ear production (kg/ha) of 196 half-sib progenies (HSP) of the maize population CMS-39 obtained from experiments carried out in four environments were used to adapt and assess the BLP method (best linear predictor) in comparison with to the selection among and within half-sib progenies (SAWHSP). The 196 HSP of the CMS-39 population developed by the National Center for Maize and Sorghum Research (CNPMS-EMBRAPA) were related through their pedigree with the recombined progenies of the previous selection cycle. The two methodologies used for the selection of the twenty best half-sib progenies, BLP and SAWHSP, led to similar expected genetic gains. Genotoxic activity of the insecticide Nuvacron (Monocrotophos) detected by the micronucleus test in bone marrow erythrocytes of mice and in CHO cells Braz. J. Genet.

The organophosphorus insecticide Nuvacron (Monocrotophos) is a very toxic agent widely utilized in Brazilian agriculture. To evaluate the clastogenic potential of this insecticide, in vivo and in vitro micronucleus (MN) assay experiments were carried out on Swiss mice and on Chinese hamster ovary (CHO) cells, respectively. Nuvacron administered at doses of 2.5 and 5.0 mg/kg induced a statistically significant increase in the frequencies of MN detected in polychromatic bone marrow erythrocytes from animals (six/group) treated ip 24 h before. Correlations of measures of percentages of white coat color, five measures of production and two measures of reproduction were obtained from 4293 first lactation Holsteins from eight Florida dairy farms. Percentages of white coat color were analyzed as recorded and transformed by an extension of Box-Cox procedures. Statistical analyses were by derivative-free restricted maximum likelihood (DFREML) with an animal model. Induction of mutations for earliness in the soybean cultivar Paraná Braz. J. Genet.
Mutation induction breeding through gamma ray seed treatment (22 krad) was used to obtain earliness in the soybean cultivar Paraná. Plants flowering earlier than Paraná were selected at the M3 generation. Eight mutant lines were isolated by further selections through several generations and tested in four yield trials carried out in 85/86 (Piracicaba) and 86/87 (Piracicaba, Sertãozinho and Assis) in São Paulo State. The following data were taken in these trials: number of days to flowering, yield, total plant height and height to pod insertion, flower color, pod pubescence and seed and plant growth type. Machado-Joseph disease (MJD) is a form of autosomal dominant spinocerebellar ataxia first described in North-American patients originating from the Portuguese islands of the Azores. Clinically this disorder is characterized by late onset progressive ataxia with associated features, such as: ophthalmoplegia, pyramidal and extrapyramidal signs and distal muscular atrophies. The causative mutation is an expansion of a CAG repeat in the coding region of the MJD1 gene. Diallel cross analysis for young plants of brachytic maize (Zea mays L.) varieties Braz. J. Genet.

Six brachytic maize varieties were crossed in a diallel mating scheme. Both varieties and crosses were grown hydroponically in a greenhouse, in randomized complete blocks with three replications in two seasons. Four brachytic double cross hybrids were used as checks. Twenty-eight days after planting, data for eight traits were taken for weights of the total plant (TPW), top plant (TOW), total roots (TRW), seminal roots (SRW), and nodal roots (NRW) and number of total roots (TRN), seminal roots (SRN), and nodal roots (NRN). Variations in egg length were observed for two populations of cryptic species of Anastrepha fraterculus (Wiedemann). The eggs of type I flies were smaller than those of type II individuals. For both types, in regard to yolk mass extrusion, four classes of embryos were detected. Class 1: embryos that extrude masses at both extremities; class 2: embryos in which extrusion occurs only at the anterior pole; class 3: embryos that eliminate mass only at the posterior pole, and class 4: embryos that do not extrude any mass. Isozyme patterns and their genetic control in three Centrosema species are described. Seven isozymatic systems (aspartate aminotransferase, glucose-6-phosphate isomerase, phosphoglucomutase, anodal peroxidase, malate dehydrogenase, 6-phosphogluconate dehydrogenase, and isocitrate dehydrogenase) were studied in 18 populations and several breeding lines of C. acutifolium, C. brasilianum and C. pubescens, using starch gel electrophoresis techniques. All systems, except glucose-6-phosphate isomerase, are described for the first time in these species.
